Hunter Doohan as Bastian Cooper / Muse: Bastian Cooper was inspired by Heather Glenn's work as a therapist to finally become his true self, which leads to him becoming Muse. With at least 60 confirmed victims, Muse is a graffiti artist and serial killer who comes into conflict with Daredevil. Muse drains his victims of their blood and uses it to paint his murals. He is killed by Heather Glenn while he is fighting Daredevil after attempting to kill her during a therapy session.

Kamar de los Reyes as White Tiger: White Tiger makes his debut in the MCU as a vigilante who uses amulets to give him his powers. He is accused of killing a police officer but Matt Murdock is able to get him acquitted, before he is shot and killed operating as White Tiger. It is the last role of the late Kamar de los Reyes.

Wilson Bethel as Benjamin Poindexter, AKA Bullseye: Benjamin Poindexter, AKA Bullseye, first appeared in Daredevil season 3 and proved to be one of the most significant threats Daredevil ever faced. He returns to Daredevil: Born Again, steps into his Bullseye persona instead, and murders Foggy. In the comics, it's Karen who Bullseye kills, marking a change for the MCU series. Bullseye is sentenced to life in prison without parole for the murder of Foggy. It is revealed that he was hired by Vanessa to kill Foggy, and he escapes prison to go murder Fisk.

Camila Rodriguez as Angela Del Toro: After being shown during Hector Ayala's trial, Angela Del Toro officially appeared in episode 4 to see Hector's body and speak with Matt Murdock. She makes her anger known about the corruption within the city and the NYPD, setting her up to eventually take over the mantle of White Tiger. Her capture by Muse prompts Matt Murdock to become Daredevil once again, possibly setting her on the path to being her own hero.

Elden Henson as Foggy Nelson: Foggy Nelson is the best friend of Matthew Murdock as well as his law partner for years, often defending clients pro bono. He is a significant aspect of Matt's life and one of the first people to learn that he's Daredevil. However, in the premiere of Daredevil: Born Again, Foggy is fatally shot by Bullseye and his death changes the lives of both Matt and Karen.

Deborah Ann Woll as Karen Page: Karen Page is one of the most significant love interests in Daredevil history, and while the original Netflix series explored that aspect of the character, she grew far beyond that, finding her own story to tell. She was a partner alongside Foggy and Matt at the Nelson, Murdock, and Page law firm. After Foggy's death, though, Karen moves across the country to San Francisco and no longer talks with Matt, though she returns to help in the finale.

Nikki M. James as Kirsten McDuffie: Kirsten McDuffie is a former assistant DA who ends up becoming Matt Murdock's new partner at Murdock & McDuffie after Foggy is killed and Karen leaves New York. She also sets Matt up with Heather Glenn.

Clark Johnson as Cherry: A former detective for the NYPD, Cherry now works as Matt Murdock's private investigator. He also knows that Matt was Daredevil after seeing him unmasked in the aftermath of the Bullseye fight. Cherry does not want Matt to come back as Daredevil.

Genneya Walton as BB Urich: BB Urich operates as a young journalist in Daredevil: Born Again, who is the niece of Ben Urich, a significant character who was killed by Wilson Fisk in the original series. She is incredibly resourceful and has been seen using Daniel Blake to get some information about Wilson Fisk's plans. She knows that Fisk is connected to her uncle's death.

Michael Gandolfini as Daniel Blake: The character is a member of Wilson Fisk's mayoral staff and could end up as his protégé. He impresses Fisk immediately with his enthusiasm for his role. His "friendship" with BB Urich got him to spill important details about what they are doing, and vaguely threatens her when she continues to push against Fisk. He is appointed Deputy Mayor of Communications by Fisk.

Arty Froushan as Buck Cashman: Like Wesley in the original series, Buck Cashman operates as Wilson Fisk's right-hand man, setting up therapy sessions led by Heather Glenn for Wilson and Vanessa while doing various other jobs that he is asked to do, which includes helping run his criminal operation and killing some people.

Michael Gaston as Commissioner Gallo: The character found himself entering into a reluctant partnership with Wilson Fisk after he was blackmailed following his attempt to step down as the commissioner of the NYPD. He helps Wilson Fisk set up his anti-vigilante task force before being taken out by Fisk in the most graphic death in MCU history.

Hamish Allan-Headley as Officer Powell: A corrupt police officer working for the NYPD, Officer Powell was one of the men that Hector Ayala fought on the subway platform. After finding the witness, Powell goes to kill him, but Matt Murdock takes him down during their fight. While Matt thought he knew who killed Hector, it is revealed that he didn't know. He is part of Fisk's anti-vigilante task force, and seemingly no longer takes any orders from Commissioner Gallo.

Jeremy Earl as Cole North: He is another member of Wilson Fisk's anti-vigilante task force. Though he hasn't been seen much yet in the series, Cole North is more important than some might realize, indicating that his storyline seems to only be beginning, especially as he was eyeing BB Urich talking to Gallo at Fisk's party. It's also possible that he died during the explosion at Matt's apartment in the finale, where it was revealed that he was the one who killed Hector.

Zabryna Guevara as Sheila Rivera: She operates as Wilson Fisk's mayoral campaign director and helps out with his decisions in politics. She is often at odds with how he conducts himself in office. By the end of the season, she is complicit in Kingpin's dealings as she fears for her life.

Ruibo Qian as Angie Kim: She is a detective and former colleague of Cherry's, and is seen in the first episode discussing with him his retirement right before Bullseye kills multiple people at Josie's. She acts as the hostage negotiator during the St. Patrick's Day bank robbery, and is the one to discover that Muse is Bastian Cooper.

Mohan Kapur as Yusuf Khan: Mohan Kapur returns to the MCU as he reprises his role as Yusuf Khan, Kamala Khan's father. He comes across Matt Murdock, who is applying for a loan for his firm, and the two of them get caught up in a bank robbery, working together to stop it. He mentions Kamala and her superhero persona, Ms. Marvel, multiple times.

Tony Dalton as Jacques "Jack" Duquesne: Tony Dalton also comes back as Jack Duquesne, a talented swordsman who initially premiered in Hawkeye, and was engaged to Kate Bishop's mother. He is seen engaged in a tense conversation with Wilson Fisk about his stance on vigilantes in New York City. He is being held captive at the end of the season.

Lou Taylor Pucci as Adam: When Wilson Fisk was absent, his wife Vanessa began an affair with Adam. Of course, Wilson Fisk didn't take to that too well, and despite telling Vanessa he would be safe, but Fisk kept Adam locked away as a prisoner, where he was able to physically hurt him throughout Daredevil: Born Again, until he gave Vanessa the choice to kill him or set him free, and she chose to kill him.
